【摘要】 目的 研究 As2 O3对肝癌细胞株 HL E细胞毒和诱导凋亡作用。方法 应用 MTT染色 ,流式细胞仪和电子显微镜观察 As2 O3对 HL E细胞株的诱导凋亡作用和形态学改变 ,应用共聚焦激光扫描显微镜 (CL SM)测试肝癌细胞内 [Ca2 + ]i变化。结果  As2 O3对肝癌细胞株 HL E具有诱导凋亡作用 ,CL SM检测肝癌细胞内 [Ca2 + ]i高浓度 As2 O3作用下 4 8h达最高峰 ;在低浓度下 96 h达高峰。结论  As2 O3对肝癌细胞具有较强的细胞毒和诱导细胞凋亡的作用。

【Abstract】 Objective To study the effects of cytotoxicity and induction of apoptosis by arsenic trioxide(As 2O 3) on hepatoma cell line HLE.Methods The effect of cytotoxicity by As 2O 3 on hepatoma cell lines HLE was determined by MTT assay, apoptosis and morphologic changes of HLE induced by As 2O 3 by electronic microscope and flow cytometry. The intracellular Ca 2+ concentration was detected by confcal Laser scanning microscope(CLSM).Results As 2O 3 could induced apoptosis in hepatoma cell lines remarkably, HLE cells exhibited typical morphologic features of apoptosis. The intracellular Ca 2+ concentration reached peak 48 hours after treatment with high concentration of As 2O 3, and 96 hours after treatment with low concentration of As 2O 3. Conclusions As 2O 3 exhibited significantly cytotoxicity and induction of apoptosis on hepatoma cell lines HLE.As 2O 3 might be promising in the treatment of hepatic carcinoma, and be worthy to be further studied.
