动脉球囊损伤丝裂素活化蛋白激酶基因表达的探讨

【摘要】 目的 :探讨丝裂素活化蛋白激酶 (MAPK)在内膜增殖过程中基因蛋白表达的变化。方法 :4 0只雄性Wistar大鼠随机分成 4组。损伤组行胸腹主动脉球囊损伤 ,术后 3d、7d和 14 d取材 (n=10 ) ;对照组 (n=10 )不进行球囊损伤。应用蛋白印迹杂交及逆转录聚合酶链反应方法观察主动脉损伤后内膜增殖过程中 MAPK表达的变化。结果 :动脉球囊损伤后 3d、7d和 14 d,MAPK活性、蛋白和基因表达均高于对照组 ,以 3d〔 MAPK活性 :(17.32± 2 .17) pm ol· mg- 1 · m in- 1 ;MAPK蛋白水平 :ERK1为 194 .7± 8.6 ,ERK2为 175 .8±7.9;MAPK基因表达 :ERK1为 1.15± 0 .2 1,ERK2为 1.13± 0 .14〕表达最为明显。结论 :MAPK在动脉损伤后异常表达 ,可能与平滑肌细胞迁移、新生内膜形成及血管重塑有关。

【Abstract】 Objective: To observe the changes in mitogenactivated protein kinase (MAPK) activity and gene expression after coronary artery balloon injury in rat. Methods: Forty Wistar rats were randomly divided into control group (without coronary artery balloon injury), and 3, 7 and 14 days after coronary artery balloon injury groups ( n =10, respectively). The activity of MAPK was measured by biochemical method and the gene expression was examined by reverse transcriptionpolymerase chain reaction (RTPCR). Results: MAPK activity and gene expression of MAPK in the coronary artery balloon injury in rat after 3, 7 and 14 days were significantly higher than that of normal control, especially 3 days after injury 〔MAPK activity:(17 32± 2 17) pmol·mg -1 ·min -1 ; MAPK protein: ERK1 was 194 7±8 6, ERK2 was 175 8±7 9; MAPK gene expression: ERK1 was 1 15±0 21, ERK2 was 1 13±0 14〕. Conclusion: The increase activity and mRNA of MAPK may be involved in the restenosis process after coronary balloon injury.
