大鼠单侧输尿管梗阻后黏附分子与树突状细胞表达分布及抗黏附干预的影响

【摘要】 目的探讨黏附分子与树突状细胞 (DC)在大鼠单侧输尿管梗阻后肾组织中的表达和分布 ,以及抗P -选择素单抗的抗黏附调抑作用。方法建立大鼠单侧输尿管梗阻 (UUO)模型 ,随机将大鼠分为手术组 (n=18)和抗P -选择素单抗治疗组 (n=18) ,按不同梗阻时间 (1、3、7d)再分为3组 ,另设假手术组 (n=6)作为对照。分别采用免疫组化LSAB法和免疫双染与荧光图像分析法 ,观察P -选择素、ICAM -1及DC在肾组织中表达和分布变化。结果大鼠UUO第1d起即见P -选择素以肾小管上皮细胞为主在肾小管间质中表达上调 ,伴随CD1a+ CD80+ DC在以肾间质为主部位的浸润。第3dP -选择素明显表达 ,ICAM -1以肾血管为主表达上调 ,且CD1a+ CD80+ DC显著增多。至第7dP -选择素表达遂下调 ,ICAM -1表达明显增强 ,而CD1a+ CD80+ DC在肾间质分布聚集达峰。经抗P -选择素单抗处理后 ,大鼠肾组织P -选择素、ICAM -1表达受到抑制 ,CD1a+ CD80+ DC分布聚集减少 ,且肾小管间质病理损伤减轻。结论P-选择素等黏附分子介导树突状细胞迁移聚集 ,可能参与了肾小管间质纤维化早期病理损伤过程 ,抗P -选择素单抗对此具有抗黏附调抑和肾脏保护作用。

【Abstract】 Objective To explore the expression of the adhesive molecules and distribution of dendritic cells(DCs)in rat kidney with unilateral ureteral obstruction as well as the preventive effect of anti-P-selectin lectin-EGF domain monoclonal antibody(PsL-EGFmAb).Methods Rat models of unilateral ureteral obstruction were established,which were divided into sham group(n=6)、untreated group(n=18)and treated group with PsL-EGFmAb(n=18).DCs were analyzed with Axioplan2microscopy,while P-selectin and ICAM-1were analysed by immunohistochemistry LSAB,the change of renal pathology was observed at the same time.Results P-selectin was present in renal tubular epithelial cell after unilateral ureteral obstruction on day1,and CD1a + CD80 + DC was also found in renal interstitium;P-selectin and DCs presented were increased evidently on the3 rd day,and the expression of ICAM-1was up-regulated in renal vessel.The expression of P-selectin was deˉcreased,but ICAM-1was increased and the accumulation of DCs was the most in renal interstitium on the7 th day.These changes became less conspicuous in rat treated with PsL-EGFmAb.Conclusions DCs immigration and accumulation may play an important role in early tubulointerstitial lesions.PsL-EGFmAb has anti-adhesive effect on it.
