香烟尘粒对血管平滑肌细胞生长的影响
Effects of Dimethylsulfoxide(DMSO)-soluble Particles from Cigarette Smoke on Vascular Smooth Muscle Cell
【摘要】 目的观察二甲基亚砜溶解的香烟尘粒 (DSP)对血管平滑肌细胞(VSMC)生长的影响。方法以兔主动脉血管平滑肌细胞为实验对象 ,将1、2、3、4、8μl·ml -1剂量DSP加入培养基中。采用四氮唑盐 (MTT)比色试验和细胞蛋白测定评价DSP对VSMC生长的影响 ;透射电子显微镜 (TEM)观察细胞超微结构变化 ;流式细胞仪 (FCM)检测细胞凋亡情况。结果1、2μl·ml-1DSP可刺激VSMC增殖 (P<0.01) ,4、8μl·ml -1DSP能抑制VSMC增殖 (P<0.05,P<0.01) ,减少细胞总蛋白 (P<0.05,P<0.01)。电镜下 ,2、4μl·ml-1 DSP组均有典型的细胞凋亡形态学变化。2μl·ml-1 DSP组凋亡率为(10.2±3.2) % ,4μl·ml-1DSP组凋亡率为 (29.2±8.7) % ,均较对照组显著增高 (P<0.05,P<0.01)。结论DSP既可刺激VSMC增殖 ,又可促进VSMC凋亡 ,吸烟参与动脉粥样硬化病理过程的机制 ,可能与其影响VSMC增殖和凋亡两者的平衡有关。
【Abstract】 Objective To investigate the effects of Dimethylsulfoxide(DMSO)_soluble particles from cigarette smoke(DSP)on vascular smooth muscle cell(VSMC).Methods Using cellular total protein and MTT assay to evaluate the effects of DSP on VSMC growth.Transmission electron microscopy(TEM)was used to observe the ultrastructural changes of VSMC.Apoptosis of VSMC was analyzed by flow cytometer(FCM).Results1l·ml -1 ,2μl.ml -1 DSP stimulated VSMC proliferation(P<0.01).4μl.ml -1 and8μl·ml -1 DSP inhibited VSMC proliferation(P<0.05,P<0.01)and had distinctive toxic effects on vascular smooth muscle cell by reducing total cellular protein(P<0.05,P<0.01).TEMshowed that2μl·ml -1 and4μl·ml -1 DSP groups exhibited morphologic typical features of apoptosis,including cell shrinkage,nuclear condensation,and forˉmation of apoptotic bodies,and marked subdiploid peaks were observed by FCM.The apoptotic rate of the cells in2μl·ml -1 and4μl·ml -1 DSP were(10.2±3.2)%and(29.2±8.7)%respectively.Conclusions DSP can not only stimulate vascular smooth muscle cells proliferation,but cause their apoptosis.The results might be related to atherogenesis.
