高粱3197A雄性不育系热激处理后代的可育性遗传

【摘要】 对高粱细胞质雄性不育系 3197A经热激处理后获得的可育结实后代进行了连续多代自交及回交观察 ,结果表明 ,由热激诱导的可育性变化不能稳定遗传。从H2 代开始 ,其可育率逐代降低。到H5代 ,分离出的可育株数仅占观察植株总数的 1.9%。H5代可育株与不育株花药总RNA与HSP70 热激蛋白基因探针Northern杂交结果表明 ,H3A可育株均能与HSP70 基因探针杂交 ,显示较强的杂交信号。而其不育株 ,有的完全不能与HSP70 基因探针杂交 ,有的显示微弱的杂交信号。说明 3197A雄性不育系热激可育后代的遗传稳定性与HSP70 基因的转录状况有关。

【Abstract】 Fertile seeds from male sterile line of sorghum 3197A(3A) were obtained after heat shock at 43-45℃ in our previous experiment. To study the genetic character of the above progenies(H3A), we observed their fertility change by inbreeding and backcrossing. The total RNA of another in pollen mother cell(PMC) of the fertile and sterile plants from H3A was extracted, and Northern blotting hybridization was performed to the probe of HSP 70 gene. The results are as follows:(1)The progenies in self crossing H 2-H 5 of H3A showed fertility segregation (Table 1),and the proportion of male fertile plants was gradually reducing(100% down to 1.9%) from H 1 to H 5.It shows that the male fertility of H3A could not be stably inherited, and there is the possibility of complete loss of fertility.(2)The results of backcrossing of the male sterile plants from H3A to maintainer line of sorghum 3197B suggested that the sterility of H3A segregated progenies was not maintained by maintainer line 3197B(Table 2).It indicated that the sterility of the male sterile plants of H3A differed from those of the male sterile line 3197A.(3)Northern blotting analysis showed that the male fertile plants from H3A had strong transcription activity of HSP 70 gene, and some sterile plants from H3A did not give hybridization signal while some gave weak hybridization signal(Fig.2). It shows that the fertility restoration of H3A is related to the expression of HSP 70 gene.
