静注克洛素隆在绵羊体内的药代动力学研究

【摘要】 应用 YWG- C1 8色谱柱 (2 5 0 m m× 4 .6 mm,5 μm) ,Waters TM 4 86型可调波长紫外检测器 (检测波长 2 6 5 nm) ;0 .0 1 M磷酸钾 (p H=7)∶乙腈 (3∶ 1 )为流动相 ;含量测定采用标准曲线法 ,建立了 RP- HPL C法检测绵羊血浆中克洛素隆含量的方法。同时对绵羊单剂量静脉注射 7mg/ kg克洛素隆的药代动力学进行了研究。血药浓度在 0 .0 0 5~ 2 .0 μg/ ml及 2 .0~ 5 0 .0 μg/ ml范围呈良好线性关系 (r=0 .994 1、0 .9970 ) ,平均回收率93.2 % ,血药最低检测浓度 0 .0 0 5 μg/ ml,日内日间变异系数分别小于 1 0 %、1 1 %。血药浓度结果经 MCPKP药代动力学统计软件处理 ,体内药物运转符合三室开放模型 ,主要药物动力学参数 :t1 /2γ=1 4 .1 4 h,Kel=0 .32 6 0 h- 1 ,AU C=1 5 3.95 m g/ l· h,fc=0 .0 5 5。结果表明克洛素隆静脉单次给药后体内分布较为广泛 ,持续作用时间长 ,主要通过肾脏排泄

【Abstract】 A RP-HPLC method was used for the analysis and pharmacokinetics studies of clorsulon in sheep plasma was established.The RP-HPLC was equiped with a YWG-C 18 column(250mm×4 6mm,5μm),Waters Tm 486 ultraviolet detector with wavelength fixed at 265nm. The mobilephase was 0 01 M sodium phosphate(pH=7 0) -acetonitrile=3∶1.Standard curve mothod was applied for determinating parent drug concentration.The results indicated that clorsulon within 0.005~2 0 μg/ml and2 0~50 μg/ml range had a good linear relationship(r=0 9941,0 9970).The limit of detection(LOD) was0 005 μg/ml,the average recovery of the method was 93 2%,the RSD% of intra-day and inter-day assays were less than 10%, 11% individually. Normal sheeps were given Clorsulon 7 mg/kg via a single i.v.administration,plasma drug concentration-time courses was found to be fitted to a three cogent open model and its phanmrcokinetic parameters was listed as follows, t 1/2γ =14 14h, Kel=0 3260h -1 ,AUC=153 95 mg/l·h, f c =0 055.Results showed that Clorsulon following a single intravenous administration was distributed widely, eliminated slowly,mainly excreted through kidney.
