反相高效液相色谱法测定饲料中喹乙醇的含量
Determination of Olaquindox in Feeds with PR-HPLC
【摘要】 建立了反相高效液相色谱法测定饲料级喹乙醇、预混合饲料和配合饲料中喹乙醇含量的方法。色谱柱为C1 8,1 5 0× 4.6mm ,5 μm ;流动相为甲醇 +水 ( 1 5∶85 ,V/V) ;检测波长为 2 60nm ;柱温为室温。结果表明 :在 0~ 1 83.2 6μg/mL之间呈现良好的线性关系 ,相关系数r2 =1 .0 ;3个不同浓度样品加样回收率分别为 96.1 4%、98.61 %和 99.42 %。本方法简便 ,快速 ,准确度和精密度较高 ,适用于饲料中喹乙醇含量的测定
【Abstract】 A high performance liquid chromatographic(HPLC) method was developed for determination of olaquindox in feeds. Olaquindox was extracted with 5% methanol solution, shaked and filtered. An external standardization method was adapted for quantitative. A C 18 stainless steel column and UV detector at 260 nm wavelength were chosen for the HPLC. Methanol solution(15%, pH 2.2) was used as the mobile phase. The average recovery of olaquindox in feeds at three different levels was 96.14%, 98.61% and 99.42% respectively. A good linear response was obtained for olaquindox standard from 0~183 μg/mL, and the correlation coefficient was 1.0.
