重组杆状病毒表达的2株传染性支气管炎病毒S1蛋白的免疫原性评价
Immunogenicity Evaluaton of S1 Glycoproteins from Infectious Bronchitis Virus Isolates JS/95/03 and SD/97/01 Expressed by Recombinant Baculoviruses
【摘要】 利用 Bac- to- Bac杆状病毒表达系统构建了 2株重组杆状病毒 r Ac JS95 0 3S1和 r Ac SD970 1S1,分别表达了 2株致病性不同的传染性支气管炎病毒的 S1基因。用感染重组杆状病毒的昆虫细胞裂解液对 2周龄的伊莎公雏进行免疫 ,根据鸡免疫后的抗体水平动态变化和攻毒后鸡肾脏和气管的保护力判定它们的免疫原性。结果显示 ,重组杆状病毒表达的 IBV S1蛋白可以诱导抗体产生和免疫保护反应 ,2毒株间也存在着一定程度的交叉免疫保护反应。
【Abstract】 Two recombinant baculoviruses rAcJS9503S1 and r AcSD9701S1 expressing S1 genes of infectious bronchitis virus isolates JS/95/03 and SD/97/01 were constructed by using Bac-to-Bac baculovirus expression syste m.The lysates of insect cells infected with recombinant baculoviruses were used to immune two-week-old commercial ISA cockerels.The immunogenicity of recombin ant S1 glycoproteins(rS1) were evaluated based on the antibody response detected by enzyme-linked immunosorbent assay(ELISA) after two immunizations and the pr otection tested by re-isolation of viruses from the kidneys and tracheas of vac cinated chickens after challenge with JS/95/03 or SD/97/01.The results indicated that S1 gene product expressed by recombinant baculoviruses could induce antibo dy responses as well as protective immune responses and a certain degree of cros s-protection was occurred between the two IBV isolates.
