银杏悬浮细胞叶绿体的分化与黄酮类产物积累

【摘要】 研究了来源于银杏GinkgobilobaL 胚和萌发30d幼苗茎的悬浮细胞在培养过程中,细胞叶绿体的分化和黄酮类化合物累积的关系。研究表明,有叶绿体分化细胞中的黄酮类化合物的质量分数高于无叶绿体分化的细胞。光照培养、增加培养基中的微量元素、添加低浓度的TDZ等促进细胞中叶绿体分化的方法都有利于悬浮细胞中黄酮类化合物的积累,选育的绿色细胞悬浮培养物中黄酮类化合物质量分数最高可达细胞干质量的0 798%。

【Abstract】 The relationship of chloroplast’s differentiation and flavonoids’ accumulation in suspension cell derived from embryo and stem of 30 days old seedings of Ginkgo biloba L. was investigated. The flavonoids content in suspension cells which had large amount of chloroplasts was higher than that of the cell with lower chloroplast’s differentiation. The culture condition such as cultured in light, increasing microelement consistency in medium, addition of thidiazuron at 0.1 μmol/L, which improved the chloroplast differentiation could also increased the flavonoids production.The maximal flavonoids content in selected green cell line was 7.98 mg/g. It was conjectured that the chloroplast maybe one of the site for flavonoids synthesis in the cell of Ginkgo biloba L.
