咪唑斯汀对花生四烯酸人皮肤炎症模型的抗炎作用研究

【摘要】 目的 :探讨咪唑斯汀对花生四烯酸 (AA)人体皮肤炎症模型的抗炎作用。方法 :以 4 %、2 0 %和 4 0 %浓度的AA在 30名健康志愿者前臂屈侧做点刺试验 ,并在其服用抗过敏药 (咪唑斯汀 /氯雷他定 )前 1h及服药后 2h进行AA点刺试验 (部分志愿者于服药后 3h再行AA点刺试验 ) ,分别记录点刺试验 30min后的红斑面积和风团面积。用氯雷他定作为对照组。结果 :2 0 %AA人体皮肤点刺试验中 ,服用咪唑斯汀前后红斑面积及风团面积变化显著 (P <0 .0 5 ,P <0 .0 1) ,4 %、4 0 %AA点刺试验中 ,咪唑斯汀组用药前后风团面积存在显著性差异(P <0 .0 5 ) ,显著优于氯雷他定组 (P <0 .0 5 ,P <0 .0 1) ;咪唑斯汀 2h抗炎效果略优于 3h ,但无统计学差异 (P>0 .0 5 )。结论 :咪唑斯汀具有明显的拮抗AA引起的人体皮肤炎症反应作用。

【Abstract】 Objective:To study the anti-inflammatory effect of mizolastine on human skin inflammation induced by arachidonic acid(AA). Methods: 30 healthy volunteers were divided into two groups randomly. They were administrated either a single dose of mizolastine 10 mg or loratadine 10 .mg in a double-blind manner. Prick tests were performed by 8μl of prepared AA on inner side of forearms. Before antihistamines were given orally 1h and after 2h and 3 hr, the sizes of erythema and wheal were measured separately 30 min after pricking. Results: Both the sizes of flush areas and wheal areas at the AA concentration of 20%, before and after mizolastine administration were significantly different ( P <0.05, P <0.01). The sizes of wheal areas AA concentration of 40% and 4%, before and after mizolastine administration were significantly different ( P <0.05). There was no difference between the anti-inflammatory effects of mizolastine after 2h and 3h ( P >0.05). Loratadine showed no significant inhibition on the inflammatory action of 4%AA and 20%AA and 40% ( P >0.05). Conclusion: Mizolastine had a strong inhibitory effect on the AA-induced inflammation of human skin.
