【摘要】 目的　将获得的抗日本血吸虫循环抗原的单链抗体进行抗原定位和用于诊断。方法　用间接荧光抗体试验(IFA)对 2株可溶性单链抗体所针对的抗原进行定位 ;将可溶性单链抗体进行生物素标记后 ,用于检测急、慢性血吸虫病人血清标本。结果　两个特异性单链抗体分别于日本血吸虫成虫的肠管和生殖系统呈现明显的荧光反应。检测急性血吸虫病人血样 2 0份 ,慢性血吸虫病人血样 30份 ,正常人血样 2 0份。结果用其中 1株单链抗体检测急性病人的敏感度为 6 0 % ,慢性病人的敏感度为 36 7% ,特异度为 90 % ;以 2株单链抗体混合检测急性病人的敏感度为 75 % ,慢性病人的敏感度为 5 6 7% ,特异度为 85 %。结论　经抗体库技术制备的单链抗体可用作日本血吸虫病的诊断 ,单抗组合较单株单抗可提高免疫学检测的敏感性。更多还原

【Abstract】 Aim To locate the antigens binding single-chain variable fragments(ScFvs) against Schistosoma japonicum(Sj) CAg and investigate diagnostic effect of ScFvs Methods The antigens were located binding two soluble ScFvs with IFA Two soluble ScFvs labeled with biotin were used in asssying sera of patients with schistosomiasis Results Two ScFvs were located in the intestine and reproductive system of Sj’s adult worm Testing acute and chronic patients with one of ScFvs or two ScFvs mixed,sensitivity are 60%,36 7% and 75%,56 7% respectively Specificity are were 85%～90% Conclusions The ScFvs manufactoried through phage antibody library may be used in diagnosis of schistosomiasis Two ScFvs mixed have higher sensitivity than a ScFv in immunologic diagnosis更多还原
