金葡菌L型DNA原位核酸杂交在小鼠肿瘤及癌前病变中的表达

【摘要】 目的 探讨金葡菌L型感染后小鼠肿瘤及癌前病变中是否有金葡菌L型及其抗原或DNA。方法 应用革兰氏染色、免疫组化及原位核酸杂交方法。结果 小鼠肿瘤及癌前病变组织切片中 ,革兰氏染色金葡菌L型检出率分别为 70 0 %(7/ 10 )和 5 3 5 % (7/ 13) ;免疫组化染色 (S -P法 )抗原阳性率分别为 70 0 % (7/ 10 )和 6 1 5 % (8/ 13) ,两者结果相似 ,无显著性差异。金葡菌L型主要分布于肿瘤间质、癌巢或癌前病变的上皮细胞浆内。用金葡菌L型DNA探针原位核酸杂交 ,结果显示 5 0 0 % (5 / 10 )肿瘤细胞核内和 30 1% (4/ 13)癌前病变细胞核内有DNA阳性信号。结论 小鼠肿瘤及癌前病变中有较高的金葡菌L型感染率 ,且金葡菌L型DNA已进入小鼠肿瘤细胞核内和癌前病变的体细胞核内。提示金葡菌L型感染小鼠可能诱发肿瘤的发生。

【Abstract】 Aim To detect the positive signal of L form A in the tumor and precancerous Lesions tissues of the mice infected with Staphylococcus aureus L form(S A) Method The changes in the tissues of 10 mice with tumors and 13 tissues of mice with precancerous Lesions were observed with Gram’s staining,immunohistochemistry and hybridization in situ Results The positive rate of S A L form were 70 0%(7/10) and 53 5%(7/13) using Gram’ s staining The positive expression of S a L form’s antigens were 70 o%(7/10) and 61 5%(8/13) by immunohistochemistry(s p staining) respectively S A L form was mainly distributed in the interstitium,nest of tumor tissues,or in the plasma of epithelial cells of precancerous Lesions Hybridization in situ showed the presence of S A L form DNA signal in the nucleus of tumor and precancerous Lesions of tested mice The positive signal were 50 0%(5/10) and 30 1%(4/13) respectively Conclusion The infection rates of S Aureus L form in tumor and precancerous Lesions tissues of tested mice were high The DNA fragments of S Aureus L form may integrate with the nucleus of mice with tumor and precancerous Lesions It suggestes that tumorigenesis may be relative to S A L form infection
