反义寡核苷酸抑制端粒酶活性对鼻咽癌细胞癌基因表达的影响

【摘要】 目的 :检测针对端粒酶的反义寡核苷酸 (AS -ODN)对鼻咽癌HNE - 1细胞株bcl- 2及c -myc基因表达的影响。方法 :用PCR -ELISA法检测细胞内端粒酶活性。用免疫细胞化学法检测bcl- 2和c -myc基因的表达。 结果 :鼻咽癌细胞株HNE - 1与 2 0 μmol/L反义寡核苷酸共同孵育 1天 ,对其端粒酶活性抑制率为 6 1.6 %。而同浓度正义寡核苷酸 (S -ODN)对细胞端粒酶活性无抑制作用。 2 0 μmol/L反义寡核苷酸作用 2天后鼻咽癌细胞c -myc基因表达HSCORE评分由 3.6 5下降为 1.98,而bcl- 2基因表达HSCORE评分无明显变化。结论 :该反义寡核苷酸可抑制鼻咽癌细胞端粒酶活性 ,此作用有序列特异性。该反义寡核苷酸抑制了鼻咽癌细胞c-myc基因的表达 ,而对bcl- 2基因表达无明显影响。

【Abstract】 Objective: To investigate the influence of AS-ODN targeting against telomerase in the expression of bcl-2 gene and c-myc gene to nasopharyngeal carcinoma cells. Methods: Telomerase activity of HNE-1 cell strain was analysed by PCR-ELISA technique.The expressions of bcl-2 gene and c-myc gene were detected by immunocytochemical method. Results: HNE-1 cells were incubated with 20μ mol/L AS-ODN for 1 day,the inhibition rate of telomerase activity was 61.6% . The S-ODN did not inhibit telomerase activity of HNE-1 cell strain at the same concentration . The score of HSCORE to c-myc gene was turned down from 3.65 to l.98 when HNE-1 cells were incubated with 20μmol/L AS-ODN for 2 days,while it did not change significantly to bcl-2 gene at the same time. Conclusion: The AS-ODN inhibits telomerase activity of HNE-1 cell strain.This procedure is sequential specific.The AS-ODN inhibits the expression of c-myc gene,but has no significant influence in the expression of bcl-2 gene.
