抗坏血酸对糖尿病大鼠肾脏PKC-β1活化的影响
The effects of ascorbic acid on activity of PKC - in rats with streptozotocin - induced diabetes
【摘要】 目的:探讨抗坏血酸对糖尿病大鼠肾脏蛋白激酶C-β1(PKC-β1)活化的影响。方法:Wistar大鼠随机分为3组,正常对照组、糖尿病组和糖尿病治疗组。实验第9周测定血浆和肾脏抗坏血酸水平,免疫荧光检测和激光共聚焦显微镜观察PKC-β1活化水平。结果:与正常对照组相比,糖尿病组血浆、肾脏抗坏血酸水平显著降低(P<0.05);肾脏PKC-β1活化水平显著增加(P>0.05);糖尿病治疗组8周后血浆、肾脏抗坏血酸水平明显增高,肾脏PKC-β1活化水平显著下降,与正常对照组相比差异尤显著性(P>0.05)。结论:糖尿病时肾脏PKC-β1活化增加可能与体内抗坏血酸水平降低有关,补充抗坏血酸可抑制糖尿病大鼠肾脏PKC-β1活化。
【Abstract】 Objective :To study the effects of ascorbic acid(vitamin C) on PKC -β1 activity in kidneys of diabetic rats. Methods :30 Wistar rats were randomly divided into three groups: Control group (group C), streptozotocin - induced diabetic rats group(group 13) and diabetic rats treated with ascorbic acid 90mg/(kg·d)(group DT). The concentration of ascorbic acid in plasma and kidney were measured at 9 week. The activity of PKC - β1 was observed by immunofluorescence technique and laser confocal microscope. Results : Compared with group C,the level of ascorbic acid in the plasma and kidney of group D decreased significantly (P<0.05) ,and the activity of PKC - β1 was markedly increased. After 8 weeks of treatment with ascorbic acid, the PKC - β1 activity was nearly normalized and the level of ascorbic acid in the plasma and kidney was increased significantly. Conclusion : This finding suggests that the increased PKC - ft activity in kidneys of diabetic rats may be associated with lower levels of ascorbic acid. Administration of ascorbic acid can inhibit the PKC- β1 activation in kidneys of diabetic rats.
