免疫磁性分选骨髓干细胞亚群Thy-1low Lin- Sca-1+

【摘要】 目的 采用免疫磁性细胞分选系统 (MACS)分离纯化骨髓干细胞亚群Thy 1lowLin Sca 1+ 。方法 收集小鼠股骨骨髓细胞 ,用MACS系统 ,通过三步分选步骤选择Thy 1lowSca 1+ 细胞 ,去除Lin+ 细胞 ,得骨髓干细胞亚群Thy 1lowLin Sca 1+ ,流式细胞仪分析其纯度 ,计算回收率。结果 分选出的Thy 1lowLin Sca 1+ 细胞纯度和回收率分别为 72 .3 6% ,82 .43 % ,达到MACS分选CD3 4+ 细胞水平。结论 MACS系统能有效分选骨髓干细胞亚群Thy 1lowLin Sca 1+ ,纯度和回收率高。

【Abstract】 Objective To isolate and purify Thy 1 low Lin Sca 1 + bone marrow stem subset cells by magnetic activated cell sorting(MACS). Methods Thy 1 low Lin Sca 1 + cells from mouse bone marrow were collected through three processes by MACS.After Lin + cells were removed,Thy 1 low Lin Sca 1 + bone marrow stem cell subsets were harvasted.The purity of the cells was analysed by FACS and the reclaimation rate was counted. Results The purity and reclaimation rate of Thy 1 low Lin Sca 1 + cells were 72.36% and 82.43% respectively, which equaled to the level of isolation and purification of CD34 + cells by MACS. Conclusions It is effective to isolate and purify Thy 1 low Lin Sca 1 + bone marrow stem cell subsets by MACS, and the purity and reclaimation rate of the cells are high.
