调心方对氧化损伤型类AD大鼠脑组织兴奋性氨基酸毒性的影响

【摘要】 目的 探讨调心方对氧化损伤型类 AD大鼠兴奋性氨基酸递质——谷氨酸 (Glu)含量及其受体 (NMDAR)之 NR1 a、NR2 a m RNA水平的影响。方法 采用二羟延胡索酸 (DHF)加 Fe Cl3- ADP左侧脑室注射法 ,造成氧化损伤型类 AD大鼠模型 ;HPLC荧光法测定鼠脑氨基酸类神经递质含量 ;RT- PCR法检测鼠脑 NR1 a、NR2 a m RNA水平变化 ,以及调心方对上述各项指标的作用。结果 与对照组比较 ,AD模型鼠脑组织中的Glu含量及 NR1 a、NR2 a m RNA水平显著增高 ,调心方对此有明显的下调作用。结论 调心方能够通过下调 AD鼠脑中 Glu含量及其受体之 NR1 a及 NR2 a m RNA水平 ,解除兴奋性氨基酸毒性 ,以达到防治 AD的目的。

【Abstract】 Objective To observe the effect of the Heart Benefiting Recipe(HBR) on the excitatory amino acid transmitter glutamic acid(Glu) and its receptor NMDAR(NR1a、NR2a) mRNA level of brain in the oxidative damaged AD rat model Methods The rats were injected with the oxygen free radical generation system (DHF FeCl3 ADP) in left ventricles of the brain and induced oxidative damaged AD rat model Using HPLC to determine the contents of amino acid in brain The level of NR1a and NR2a mRNA were observed by RT PCR The regulating effects of HBR on above indices were also studied Results Compared with the normal group, the contents of Glu and the levels of NR1a and NR2a mRNA in AD rats model were increased obviously HBR, however, could decrease that of the AD rats model significantly Conclusions HBR can remove the toxicity of the excitatory amino acid through decreasing the contents of Glu and the levels of NR1a and NR2a mRNA in AD rats model, thus attaining the purpose of prevention and cure AD
