博宁霉素对银屑病试验模型的影响

【摘要】 目的考察博宁霉素对银屑病试验模型的影响。方法1.①博宁霉素分别以5和10mg/kg的剂量,对雌激素期的小鼠腹腔注射,每日一次,连续3d;②分别用含0.1%和0.05%的博宁霉素软膏涂抹雌性小鼠阴道,每日一次,连续3d,考察阴道上皮基底细胞的有丝分裂指数。2.①博宁霉素分别以5和10mg/kg的剂量对小鼠腹腔注射,每日一次,连续15d;②分别用含0.1%和0.05%的博宁霉素软膏涂抹小鼠尾部,连续15d。考察小鼠尾表皮厚度和颗粒层形成的情况。3.分别以1000、100、10、1.0、0.1、0.01μg/ml的博宁霉素作用于小鼠脾细胞悬液,考察T淋巴细胞的增殖程度。结果博宁霉素对雌激素期小鼠阴道上皮有丝分裂有明显的抑制作用,对小鼠尾部表皮增厚和颗粒形成具有明显的促进作用,对T淋巴细胞增殖具明显抑制作用且呈剂量依赖性。结论博宁霉素具有改善和逆转银屑病病理模型的作用。

【Abstract】 Objective To observe the effects of boningmycin on pathological models of psoriasis. Method (Ⅰ ) ① The mice in estrogen phase were treated with the doses of 5 mg/kg and 10 mg/kg of boningmycin (ip) for injection respectively once a day, in three consecutive days; ② In vitro, the vaginal of mice was swabbed with the ointment of boningmycin (0.1% and 0.05% ) once a day, in three consecutive days.The index of mitosis of vaginal epidermal cells were investigated. (Ⅱ ) ① The mice were treated with the doses of 5mg /kg and 10mg/kg of boningmycin for injection(ip) respectively, once a day, in 15 days; ② In vitro,the ointments of boningmycin (0.1% and 0.05% ) were spread on tail of mice, once a day, 15 times successively. To observe the cornification improvement and thickness changes of tail epidermis in mice. (Ⅲ ) 1000, 100, 10, 1.0, 0.1, and 0.01 mg/ml of boningmycin reacted with the supernatent of mice spleen cells in vitro,to observe the inhibition of T Lymphocyte multiplication. Results Boningmycin can inhibit the mitosis of vaginal epidermal cells of mice in estrogen phase and the multiplication of T- Lymphocyte in vitro dose- dependently.In addition,the antibiotic was able to improve the cornification of tail epidermis in mice (P< 0.01). Conclusion These results showed that boningmycin is active on the pathologic models of psoriases.
