用Y-参比法测定发酵液中林可霉素的含量

【摘要】 研究发酵液中林可霉素的分光光度测定方法。根据分光光度法测定林可霉素的原理,研究林可霉素的测定影响因素和规律,得出最佳测定波长为380nm、络合反应时间为30min、浓度为0.02mol/L的络合剂PdCl2剂最佳用量为0.5ml、林可霉素浓度在20~300μg/ml范围内吸光度A与林可霉素浓度C有线性关系。根据Y-参比法,建立了测定发酵液中林可霉素的含量的有效方法。

【Abstract】 A method for the determination of lincomycin concentration in the fermentation broth by spectrophotometry was proposed.The main factors affecting correct measurement of lincomycin concentration were studied,based on the theory of measuring lincomycin concentration by spectrophotometry.The detection wavelength was 380nm,the reaction time was 30min,and the amount of developer PdCl 2 was 0.5ml which the concentration was 0.02mol/L. Absorbance shows linear relationship to lincomycin concentration in the range of 20~ 300μ g/ml.Based on the method of Y- reference,an effective method of determination of lincomycin concentration in the fermentation broth was developed.
