冻干家兔眼角膜复水工艺及角膜全层检测

【摘要】 角膜的真空冷冻干燥是离体生物组织的冻干 ,目的是便于贮藏 ,以备后用。冻干角膜的复水是决定角膜活性的关键环节。本文以冻干的兔角膜为实验材料 ,对其复水进行研究 ,找到了合适的复水工艺。对按该工艺复水的冻干角膜进行了全层透射电镜检测 ,结果表明冻干角膜的上皮细胞层、前弹力膜、基质层、后弹力膜以及它门间的连接完好

【Abstract】 A novel technique has been successfully developed to hydrate freeze dried rabbit corneas.The hydrated rabit cornea was examined layer by layer with transmission electron microscopy(TEM) to evaluate its organic structures.As compared with TEM images of the control sample,a fresh rabbit cornea,no abnormal observation was found showing possible damage of the organic structures in both the layer connections and the five layers,including epithelial layer,bowman membrane,stroma,descemet membrane and endothelium layer.
