肺耐药蛋白、拓扑异构酶Ⅱ在肺癌中的表达

【摘要】 目的 探讨肺耐药蛋白(LRP)、DNA拓扑异构酶Ⅱ(TopoⅡ)在肺癌中的表达及临床意义。方法 应用免疫组化技术S-P法 ,对60例肺癌标本进行LRP、TopoⅡ指标检测。结果 在肺癌组织中LRP、TopoⅡ表达的阳性率分别为78.3%、83.3% ,其与性别、年龄、临床分型、部位和临床分期无关(均P>0.05)。LRP在未分化型、小细胞肺癌中表达的阳性率明显降低(均P<0.05) ,TopoⅡ在未分化型肺癌中表达的阳性率明显升高(P<0.05)。小细胞癌与鳞癌、腺癌、支气管肺泡癌比较 ,LRP的表达强度明显降低(P<0.05) ,而TopoⅡ表达强度的差别无显著性意义(P>0.05) ;LRP的表达强度随分化程度的降低而降低(P<0.05) ,而TopoⅡ的表达强度随分化程度的降低而升高(P<0.05)。 结论 LRP、TopoⅡ在未化疗过的肺癌组织中均有不同程度的高表达 ,且与肺癌的某些生物学行为有关 ,临床检测有助于对化疗药物的选择及预后的判断。

【Abstract】 Objective To investigate the expression of LRP and TopoⅡin lung cancer and to evaluate its clinical significance.Methods A total of60tissue specimens of lung cancer was examined with immuno-histochemistry SP method for LRP and Topo II proteins.Results The positive rate of LRP and TopoⅡin lung cancer tissues were78.3%and83.3%.Their expression was not related to gender,age,location and clinical stage of the disease(P>0.05).Lowexpression of LRP was more frequently associated with non-differentiated and small cell lung cancer,but expression of TopoⅡwas higher in non_differentiated lung cancer(P<0.05). The expression intensity of LRP in small cell lung cancer was lower than that in squamous cell carcinoma,ade-nocarcinoma and alveolar carcinoma,but the expression intensity of TopoⅡwas not differential significantly(P<0.05).The lower the expression intensity of LRP was,the poorer the cancer cell differentiated.On the con-trary,the expression intensity of TopoⅡwas increased with lower differentiated cancers(P<0.05).Conclusions LRP and TopoⅡwere over expressed to various extent in lung cancer,and related to the biology behavior of the cancer.Detecting expression of LRP and TopoⅡwas of benefit to the choice of chemotherapeutic drugs and the prediction of disease outcomes.
