骨髓基质细胞体外诱导成神经细胞的实验研究
Human bone marrow stromal cells were induced into neurons and glial cells in vitro
【摘要】 目的 探讨骨髓基质细胞 (BMSCs)体外诱导分化成神经细胞的可能性。 方法 以成人BMSCs为实验对象 ,用碱性成纤维生长因子促进细胞增殖 ,胶质细胞神经营养因子(GDNF)等作诱导剂 ,观察诱导前后BM SCs的变化。结果 用GDNF等诱导24h后大量的BMSCs出现了巢蛋白抗体的表达 ,诱导72h后 ,BMSCs向多角形或不规则形转变 ,并可见突起生成 ,甚至形成类似网络状结构 ;诱导72h即有神经元特异性烯醇化酶的表达 ,并出现微管相关蛋白 -2、胶质纤维酸性蛋白阳性细胞。结论 成人BMSCs在体外条件下 ,经GDNF等诱导可分化成神经细胞。
【Abstract】 Objectives To explore the possibility of human bone marrowstromal cells(BMSCs)differˉentiating into neurons and glial cells in vitro.Methods BMSCs were cultivated in a medium containing basic fibroblast growth factor(bFGF)and glial cell line-derived neurotropic factor(GDNF).Microscopic observation,immunohistochemical staining of Nestin,NSE,MAP-2and GFAP were performed.Results NSE,MAP-2and GFAP-positive cells were found after72h induction.Conclusion The BMSCs can differentiate into neurons and glial cells in vitro.
