应用RAPD分析红檵木变异类型的遗传多样性
Analysis of genetic diversity among Loropetalum chinense var. rubrum by RAPD
【摘要】 用随机扩增多态性DNA技术(RAPD)分析了27份檵木材料的遗传多样性,包括24个红檵木变异类型,1个长红檵木(檵木的另一变种)及2个原种檵木材料。共筛选了180个随机引物,其中16个引物能产生清晰的扩增带且多态性明显。共产生119条带,平均每个引物产生7.4条,其中多态性带96条,多态性比例为80.7%。对RAPD数据进行UPGMA聚类分析,结果表明:27个样本共分为5个聚群,聚群内各样本之间没有明显的共同特征,但各小聚类组内各样株基本上有相同的叶色或花色。2个檵木原种、长红檵木与8个红檵木类型形成了一个大聚群,2个檵木原种与2个红檵木小叶类型亲缘关系很近。檵木与长红檵木的距离小于檵木与红檵木之间的距离。
【Abstract】 Random amplified polymorphic DNA (RAPD) technique was applied analyzing a total of 27 Loropetalum chinense samples including 24 accessions of Loropetalum chinense var. rubrum (No.1-21, 25-27), 1 accession of L.chinense var. semper-rubrum (No.22), and 2 accessions of L. chinense (No.23-24). Sixteen out 180 random primers were selected which produced distinct bands, showing unmistakable polymorphism. A total of 119 bands were displayed, among which 96 were polymorphic, attaining a polymorphic rate of 80.7%. Unweighted pair-group mathematic analysis (UPGMA) was conducted on the basis of these RAPD data and disclosed that five groups could be distinguished with 27 accessions. It seemed that these groups did not correlate closely with leaf or flower color, but there were similarities for leaf and/or flower color in some minor groups. Two accessions of L. chinense were closely related to 2 small-leaf accessions of L. chinense var. rubrum. The distance between L. chinense and L. chinense var. semper-rubrum was shorter than that between L. chinense and L. chinense var. rubrum.
