【摘要】 鸡白细胞介素2(IL-2)是一种在机体免疫调节中具有多种功能的细胞因子。本研究将萧山鸡白细胞介素2基因克隆入原核表达载体pET-28b,获得了重组表达载体pET-28b-IL-2,将此重组表达载体转化大肠杆菌BL21,经IPTG诱导表达萧山鸡IL-2。SDS-PAGE结果表明,萧山鸡IL-2在大肠杆菌中得到了表达,以此表达产物制成油乳剂苗免疫家兔,用ELISA和Westernblotting检测兔血中的多克隆抗体,结果均为阳性。因而为进一步开展鸡IL-2的结构和功能研究奠定了基础。更多还原

【Abstract】 Interleukin2(IL2) is known to play a number of functions in the modulation of immunity. In this study the chicken IL2 gene was cloned into a prokaryon vector pET28b and the recipient cell of E.coli. BL21 was transformed with the recombinant expression vector pET28bIL2. IL2 was expressed under the induction of IPTG as identified by SDSPAGE. An emulsion was prepared with the purified IL2 and injected into rabbits. The polyclonal antibody in the rabbit serum is identified by ELISA and Western blotting.更多还原
