RNA干涉与基因功能研究进展
Progress on the study of RNA interference and gene function
【摘要】 RNA干涉是通过双链RNA的介导,在基因的转录后水平上,特异性抑制具有相应序列的基因表达,即通过双链RNA的介导特异性降解相应序列的mRNA,从而导致转录后水平的基因沉默。迄今,在真菌、植物、动物等真核生物中都发现存在这一基因沉默机制。RNA干涉对于抵抗病毒入侵、抑制转座子活动、生物体的发育和基因表达调控都有重要作用;它也为基因功能的研究开辟了一条新途径。文章综述了RNA干涉的机制及其在基因功能研究方面的最新进展。
【Abstract】 The paper reviewed recent progress on the study of mechanisims of RNA interference and gene function. RNA interference is a posttranscriptional gene silencing mechanism that dsRNA specifically suppresses the expression of a gene with a corresponding sequences. It degrades mRNA specifically and potently through the mediation of corresponding doublestranded RNA and leads to posttranscriptional gene silencing. Consequently RNA interference is observed in eukaryotic organisms including fungi, plants and animals. RNA interference plays important roles in resistance to virus invasion and inhibition of transportable elements. It also provides a vital tool in the study of gene function activity.
