双低油菜浙双72的小孢子培养与植株再生研究

【摘要】 研究了甘蓝型双低油菜品种浙双72及其F1代杂种的小孢子培养。结果表明,浙双72对小孢子培养有较好的反应能力,通过换培养液等方法,小孢子胚状体产量明显提高,达27.1胚/花蕾,而F1代杂种(浙双72×高油605)的胚状体产量为4.4胚/花蕾,胚状体的质量也有明显改善。正常胚状体经过冷击、萌发、生根,获得了大量的再生植株。对再生植株倍性检测表明,浙双72及其F1代杂种中,单倍体占56%左右,自发双单倍体(DH)约占37%,其余约7%是多倍体和嵌合体。

【Abstract】 The results of the embryogenesis and ploidy level of microspore culture from a new canola variety Zheshuang 72 (Brassica napus) and its hybrid indicated that Zheshuang 72 shown a good response to microspore culture. The production and quality of the embryo were greatly increased by using the procedure of renovation medium. Zheshuang 72 produced 27.1 embryos per flower bud while its hybrid (Zheshuang 72 × Gaoyou 605) produced 4.4 embryos per flower bud. Large amounts of regenerated plants were obtained after cold treatments to embryos, embryo germination, and rooting.Analysis of ploidy level of the regenerated plants revealed that there were about 56% of haploids, 37% of spontaneous diploids, and 7% of ployploids and chimerics in it.
