小鼠烧伤早期肝组织AP-1 DNA结合活性的变化

【摘要】 目的 观察烧伤小鼠早期肝组织AP - 1DNA结合活性的时相变化特点 ,初步探讨烧伤早期应激和炎症反应的分子机制。方法 将 36只BALB/C小鼠随机分为正常对照组和实验组 ,实验组给予背部Ⅲ度 15 %~ 2 0 %烧伤。分别于伤后 2、4、6、12、2 4h用EMSAs测定肝组织AP - 1的DNA结合活性。结果 烧伤后 2hAP - 1的DNA结合活性与正常对照组相比无显著差别 ,4h后AP - 1的DNA结合能力逐渐升高 ,12h达到最大值 ,以后逐渐恢复 ,至 2 4h仍显著高于正常对照组。结论 烧伤早期引起小鼠肝组织AP - 1的DNA结合活性显著增强。

【Abstract】 Objective To observe the changes of hepatic AP-1 DNA binding ability during the early stage in burned mice.Methods 36 BALB/C mice were divided randomly into control and burn group.After clipping the hair,15%~20% thickness third degree burn injury was produced.At 2,4,6,12,24 h after burn,the AP-1 DNA binding ability in liver were detected by electrophoretic mobility shift assays.Results After burn injury,the AP-1 DNA binding ability has no significant difference during the early stage (2 h), compared with control group. It markedly elevated at 4 h,peaked at 12 h and gradually returned to the normal level.But it was still obviously higher than control group at 24 h after burn injury.Conclusion The AP-1 DNA binding ability was markedly increased in liver in burned mice.
