阴道毛滴虫小鼠模型的建立及感染动物免疫学诊断的研究
ESTABLISHMENT OF ANIMAL (MOUSE) MODEL OF TRICHOMONAS VAGINALIS AND STUDIES ON IMMUNOLOGICAL DIAGNOSIS OF INFECTED MICE
【摘要】 目的 建立阴道毛滴虫动物模型,寻找较理想的免疫诊断方法。方法 用体外培养不同时间(10和30d)、不同虫数(1×10~6和1×10~5)以及不同保存方法(新分离虫株和液氮保存虫株)获取的阴道毛滴虫虫株分别于背部皮下感染小鼠,并动态观察感染小鼠的脓肿形成。用ELISA方法检测感染小鼠血清中CAg、CIC和Ab。结果 用体外培养10和30d的新分离虫株感染小鼠,脓肿出现率分别为75%和60%,最大脓肿面积分别为0.94和0.25cm~2。用新分离和经液氮保存的虫株感染小鼠,脓肿出现率分别为80%和25%,最大脓肿面积分别为1.00和0.09cm~2。以1×10~6虫数皮下接种35只小鼠,在感染4~32d后均可形成脓肿,其脓肿大小与脓液中活虫数有关;感染50~60d时,脓肿内只有死虫,无活动虫体。用ELISA方法测定感染小鼠抗体,感染40~60d的小鼠血清抗体滴度可高达1:640;用斑点ELISA方法检测感染小鼠血清中CAg、CIC,只有用毒力较强的虫株感染小鼠,才会出现阳性反应。结论 用1×10~6个阴道毛滴虫皮下感染小鼠可以建立较理想的动物模型,活虫在体内维持的最长时间为40d,且虫体致病与接种虫数、虫株体外培养时间和体外保存方法有关。感染小鼠的抗体滴度与感染持续时间有关;在毒力较强的虫株感染的小鼠体内,可测出CAg和CIC。
【Abstract】 Objective To establish animal modei of Trichomonas vaginalis and study on immunological diagnosis of infected mice. Methods T. vaginalis, from the different cultivation timed(10 days and 30 days) , trophozoites amount(1×106 and 1 × 105 ) and preservation methods (new separation and cryopreservation in liquid nitrogen), were inoculated into mice by subcutaneous injection respectively. Formation of abscesses in infected mice was observed at regular intervals. CAg,CIC and Ab in serum of infected mice were detected by ELISA. Results As to the mice infected by new separating trophozoites which were cultivated for 10 days and 30 days, their emergence rate and the maximum area of abscesses were respectively 75% , 60% and 0. 94 cm2 , 0. 25 cm2 , while as to the mice infected by trophozoites from new separation and cryopreservation in liquid nitrogen, their emergence rate and the maximum size were respectively 80% , 25% and l cm2 , 0. 09 cm2. 35 mice were infected by subcutaneous injection with 1 × 106 trophozoites. Subcutaneous abscesses were all found on the 4th-32ed days after infection. The area of abscess was found to be related to the a-mount of alive T. vaginalis in abscess fluid. There were only death organisms instend of alive trophozoites in abscess fluid of mice on the 50th-60th days after infection. The highest Ab titer in mice serum on the 40th-60th days after infection was 1 : 640 by ELISA. Only when mice were infected by T. vaginalis with stronger toxicity, CAg and CIC in serum of infected mice was positive reaction with Dot-ELISA. Conclusion The animal modei of T. vaginalis can be established, when mice were inoculated by subcutaneous injection with 1×106. The longest life-span of trophozoites in infected mice was 40 days. The pathogem’city of T. vaginalis was found to be related to the inoculating amount, cultivation time and preservation method of trophozoites. Ab titer has relationship with continued time of infection. CAg and CIC, in serum of mice infected by trophozoites with stronger toxicity, can be detected.
