【摘要】 目的 探讨不同时段缺血对SD大鼠骨及血管超微结构的影响。 方法 研究材料为SD大鼠骨及血管 ,进行冷藏缺血和常温缺血实验 ,缺血时间为 1、2、3、4、5、6和 8h。以 15%DMSO为低温保护剂 ,采用两步冷冻步骤 ,标本冷冻预处理温度 :骨及动脉为 -55℃ ,静脉为 -60℃。液氮保存 1周 ,复温后进行透射电镜观察。 结果 缺血 8h ,冷藏组血管内皮细胞轻度肿胀 ,胞浆线粒体嵴稀疏或破坏。骨组织与对照组无明显差别 ;常温组血管内皮细胞坏死增多 ,残留胞膜。骨细胞核呈固缩表现。 结论 冷藏能明显延长骨及血管组织的缺血耐受时间 ,可获得最大部分生物活性的组织

【Abstract】 Objective To investigate the effect of the ultrastucture of bone and vessel tissue on different ischemia duration of SD rat Method To adopt two frozon methods, the duration of ischemia cold storage and normal temperature is chemia experiment on SD rat vessel tissue was 1,2,3,4,5,6h and 8h, with 15%DMSO for low temperature protective reagent, the bone and artery tissue with -55℃ of holding temperature, the vein tissue with 60℃ of holding temperature, frozon pretreatment in liquid nitrogen were preserved for one week The tissue was sliced and studied by using TEM(transmission electrical microscope) Result The vessel ischemia 8 h, the artery and the vein endothelium cell of cold storage group were obviously dropys and to show up necrosis endothelium cell in individual, there were not obvious difference between the bone tissue and the control group The artery and vein endothelium cell of the normal temperature group were necrosis add to, the bone karyou was karyopycnosis Conclusion This study showed that to cold storage of ischemia bone and vessel tissue using in this hapel could prolong the duration of tissue endurable for ischemia, and thus to keep the most of biological activities of tissue
