【摘要】 目的 探讨香烟烟雾提取物 (CSE)对人外周血单个核细胞 (PBMC)IL 8mRNA表达的影响及其细胞内信号途径。方法 体外应用CSE与健康者PBMC共同培养 ,用RT PCR检测IL 8mRNA的表达 ,用免疫细胞化学染色法检测核因子κB(NF κB)的表达 ;并观察NF κB抑制剂和酪氨酸蛋白激酶抑制剂对CSE诱导的PBMCIL 8基因表达的影响。结果 加入CSE培养的人PBMC ,IL 8mRNA的表达增加 ,作用 2 4h最高 ,NF κB活化细胞百分比增加 ;NF κB抑制剂和酪氨酸激酶抑制剂均使CSE诱导的IL 8mRNA表达水平下降。结论 吸烟使人PBMC的IL 8mRNA表达水平显著增加 ,与NF κB和酪氨酸激酶有关。

【Abstract】 Objective To explore the effecte of cigarette smoke extract (CSE) on expression of interleukin-8 (IL-8) mRNA in human peripheral blood mononuclear cells (PBMC), and the intracellular signaling pathway involved. Methods Human PBMCs were cultured with 20% CSE. The expression of IL-8 mRNA was assayed by RT-PCR. The expression of nuclear factor-κB (NF-κB) was determined by immunocytochemical staining. The effects of the inhibitors of NF-κB and tyrosine kinase on the mRNA expression of IL-8 induced by CSE were studied. Results Exposure of human PBMCs to CSE resulted in an increase in the expression of IL-8 mRNA and activation of NF-κB. Treatment of PBMCs with NF-κB inhibitor pyrrolidine dithiocarbamate (PDTC) or tyrosine kinase inhibitor genistein inhibited IL-8 mRNA expression and NF-κB activation in response to CSE. Conclusion Cigarette smoke induced the expression of IL-8 mRNA which is associated with NF-κB and tyrosine kinase.
