幽门螺杆菌结构性群体形成的研究

【摘要】 目的 研究幽门螺杆菌 (Helicobacterpylori,Hp)由浮游状单细胞转变成结构性群体过程中蛋白质表达差异。方法 用超微结构形态观察及双向电泳技术比较浮游状幽门螺杆菌及其结构性群体全菌蛋白表达图谱的差异 ,将结构性群体形成过程中表达量显著增加的蛋白斑点 ,进行胶内酶切 ,肽混合物使用基质辅助激光解吸 电离飞行时间质谱仪进行质谱分析 ,获得 4个明确的肽质量指纹谱 ,通过蛋白质数据库进行检索分析。结果 与结构性群体形成密切相关的蛋白质为鞭毛丝帽蛋白、2 磷酸甘油酸酯脱水酶、3,4 邻苯二酚 2 丁酮 4 磷酸盐合成酶和热休克蛋白 33同类物。结论 上述蛋白的鉴定将有助于发现幽门螺杆菌新的致病因子以及抗菌靶位。

【Abstract】 Objective To study the comparative proteomics of Helicobacter pylori during conversion from the planktonic cell to the structured community. Methods The two-dimensional electrophoresis (2-DE) were employed to compare the global protein patterns between the planktonic cell and structured community. Protein spots remarkably increased in structured community, which were digested in gel by enzyme and the mass of generated peptides were measured by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). The data obtained from peptide mass fingerprinting (PMF) were used in protein database search. Results Four spots of proteins in gel were identified as flagellar filament cap, 2-phosphoglycerate dehydratase, 3,4-dihydroxy-2-butanone 4-phosphate sythase (DHBP synthase) and heat shock protein 33 homolog. Conclusion These proteins will be useful for the identification of new pathogenic factors and curative value against Helicobacter pylori.
