乌司他丁对急性胰腺炎大鼠肺局部致炎因子的影响
Effects of ulinastatin on pulmonary cytokines in rats induced by acute necrosis pancreatitis
【摘要】 目的 探讨乌司他丁 (UTI)对急性坏死性胰腺炎所诱发的肺局部炎症因子的影响及意义。方法 Wistar大鼠 72只随机分为正常对照组 ( 8只 ) ,ANP模型组 ( 3 2只 )和UTI治疗组 ( 3 2只 ) ,建立大鼠ANP模型。在术后 3、6、12和 2 4h用酶联免疫吸附法 (ELISA)检测肺组织匀浆中白细胞介素 (IL) 1β和肿瘤坏死因子 (TNF) α两种细胞因子的含量 ,同时进行氧分压 (PaO2 )和肺湿重/干重比值的测定。结果 ANP组术后 3hIL 1β和TNF α即明显升高 ,并分别于术后 6和 12h达到峰值。UTI组炎症因子升高幅度明显低于ANP组 (P <0 .0 5 ) ,且于术后 2 4h基本恢复正常。结论 UTI能明显减少ANP大鼠肺组织中IL 1β和TNF α的含量 ,减轻由细胞炎症因子介导的急性肺损伤。
【Abstract】 Objective To explore the preventive and therapeutic effects of Ulinastatin (UTI) in the treatment of acute lung injury induced by acute necrosis pancreatitis (ANP).Methods The Wistar rats were randomly divided into the normal control group ( n =8),ANP group ( n =32) and UTI-treated group ( n =32).The animal model of ANP was established by injection of 5% sodium taurocholate (1 ml/kg) into the choledochus.In the UTI-treated group,UTI (4×10 4 U/kg) was given intravenously through mesenteric vein at the operation.The animals were killed at the 3th,6th,12th and 24th h after operation respectively.The change of IL-1β and TNF-α levels in pulmonary tissue were determined by enzyme-linked immunosorbent assay.PaO 2 and the wet to dry lung weight ratio were also recorded for 24 h.Results In UTI-treated group,the levels of IL-1β and TNF-α in lung tissue were significantly lower than those in the ANP group ( P <0.05).Compared with the ANP group,the wet to dry lung weight ratios were decreased dramatically in the UTI-treated group,but PaO 2 contents were increased significantly ( P <0.05 for both).Conclusion UTI can inhibit the production of IL-1β and TNF-α in pulmonary tissue,decrease the injury severity of lung mediated by the cytokines.
