缺血对失神经肌肉超微结构及酶组织化学的影响

【摘要】 目的 研究肢体缺血对失神经支配骨骼肌组织学、超微结构及Na、K ATP酶和Ca ATP酶活性的影响。方法 选用SD大鼠 2 4只 ,建立双下肢失神经支配腓肠肌实验模型后 ,结扎右侧股动脉 ,观察肌细胞的组织学 ,超微结构及Na、K ATP酶和Ca ATP酶活性变化。结果 缺血侧肢体肌细胞直径及截面积较对照侧下降速度快 2 5 .48%及 3 2 .3 3 % (t =2 .94,P <0 .0 5 ;t=2 .88,P <0 .0 5 ) ;缺血加速肌细胞线粒体 ,肌质网退变 ;Na、K ATP酶活性术后 2周实验侧较对照侧增高5 9 .68% ,而术后 4周缺血侧较对照侧下降 17.45 % (t =2 .2 4,P <0 .0 5 ) ;Ca ATP酶活性下降速度缺血侧较对照侧分别快 2 6.42 %及 2 1.87% (t =2 .64、2 .2 4,P <0 .0 5 )。结论 缺血加速失神经支配骨骼肌萎缩的发生与发展 ,周围神经损伤合并肢体动脉损伤时 ,动脉尽可能予以全部修复。

【Abstract】 Objective To study the effect of ischemia onthe histology,ultrastructure and Na,K-ATPase and Ca-ATPase activities in denervated muscle atrophy.Methods Twenty-four SD rats were used and the model of denervated skeletal muscle in two lower limbs was made by cutting off the tibial nerve.Then the femoral artery of the right lower limb was ligated.The gastrocnemius muscle histology,ultrastructure,Na,K-ATPase and Ca-ATPase activities were observed.Results The reduction rate of muscle cell diameter and cross section in the right lower limb was faster 25.48% and 32.33% respectively than that of control side.Ischemia accelerated the degeneration of mitochondria and sarcoplasmic reticulum.The Na,K-ATPase activity rose abonormally in 2 weeks after operation.The reduction rates of Na,K-ATPase activity in the right lower limb was faster 17.45% than that of control side 4 weeks after operation.The reduction rate of Ca-ATPase activity was faster 26.42% and 21.87% respectively than that of the control side.Conclusion Ischemia accelerated the degeneration of muscle histology and enzymic histochemistry of denevated skeletal muscle.The blood vessel should be repaired as much as possible when the peripheral nerve injury accompanied by artery injury in the limb.
