生物瓣材料化学改性和内皮化的实验研究
Experimental study on chemical modification and endothelializatio n of biopresthetic heart valve materials
【摘要】 目的 探索一种有效的生物瓣材料内皮化方法 ,延长生物瓣的使用寿命。方法 新鲜牛心包片分 3组处理 :Ⅰ组单纯戊二醛 (GA)处理 ;Ⅱ组GA + 2 ,3 丁二醇改性 ;Ⅲ组GA + 2 ,3 丁二醇改性 +Ⅰ型胶原蛋白预覆 ;Ⅳ组空白玻片作对照。各组体外种植内皮细胞 ,计算种植后第 1、3、5、8天的活细胞数。种植后第 8天行第Ⅷ因子检测和扫描电镜 (SEM )观察。将内皮化的Ⅲ组牛心包片移植至猪腹主动脉 ,Ⅰ组牛心包片作对照。 30d后取出标本行第Ⅷ因子检测和SEM观察。结果 Ⅲ组牛心包片表面细胞生长最为活跃 ,在第 5、8天的细胞数明显多于Ⅰ组和Ⅱ组 (P <0 .0 5 ) ,经动脉血流冲击 30d后表面光滑 ,无钙化和血栓形成 ,有较多内皮细胞存留 ,细胞与基质间有微丝连接形成。而对照组牛心包片表面粗糙 ,小钙化形成 ,无细胞生长。结论 2 ,3 丁二醇改性和预覆Ⅰ型胶原蛋白是生物瓣材料内皮化的有效和可行的方法 ,有抗钙化、血栓形成和抗血流切应力能力 ,具有临床应用前景。
【Abstract】 Objective To study an effective way for e ndothelialization of biopresthetic heart valve material to improve its durabilit y.Methods Fresh bovine pericardiums (BPs) were divided in to three groups:groupⅠtreated with glutaraldehyde;groupⅡ modified with the 2 ,3-butanediol;group Ⅲ modified with the 2,3-butanediol and precoated with c ollagen type.The glass plates were used as control (group Ⅳ).Endothelial cell s (ECs) were seeded onto the surfaces of BPs,and counted on day 1,3,5 and 8, and identified by factor Ⅷ staining and evaluated by scanning electron microsco py (SEM) on the 8th day after seeding.Then Group Ⅲ BPs were transplanted to th e abdominal aortic wall of pigs.GroupⅠBPs were transplanted as control.On the day 30,the retrieved specimens were identified by factor Ⅷ staining and studi ed by SEM.Results On the day 5 and 8,the amount of ECs i n the groupⅢ were significantly more than in the groupⅠand groupⅡ (3.9±0.7, 4.2±0.8 vs 2.2±0.5,2.9±0.5,respectively,P<0.05).BPs of group Ⅲ were sim ilar to native heart valve.On the day 30 after implantation,the surfaces of the group Ⅲ specimens were smooth.No calcification and thrombosis occurred.SEM show ed lots of ECs remained,and spiderlike junctions between ECs and matrix formed.I n contrast,groupⅠspecimens were unsmooth,slight calcification occurred,and no c ells remained.Conclusion Endothelialization of biopresthe tic materials with the 2,3-butanediol modified and collagen typeⅠprecoated tre atment in vitro is effective,and can prevent calcification and thrombosis,and re sist the shear stress of blood stream.
