9-顺-维A酸诱导肺鳞、腺癌细胞株凋亡与CyclinD1和Rb基因表达的关系

【摘要】 目的 探讨 9 顺 维A酸 ( 9 cis retinoicacid 9 cisRA )诱导肺癌细胞株凋亡作用及其与CyclinD1、Rb基因表达的关系。 方法 体外培养肺鳞癌细胞株L78、肺腺癌细胞株PG ,随机分为两组 ,实验组加 9 cisRA使其终浓度为 5 μmol L ,对照组加入二甲亚砜使其终浓度为 0 .1% ,培养48h后用流式细胞仪技术分别检测CyclinD1、Rb基因表达率 ,同时用DNA凋亡分析法检测肿瘤细胞凋亡发生率 ,研究三者之间的相关关系。结果 L78、PG细胞实验组中细胞凋亡发生率显著增高 ,两株细胞中凋亡发生率与CyclinD1基因表达率之间负相关 (L78:r =-0 .70 7,P <0 .0 5 ;PG :r =-0 .785 ,P <0 .0 1) ,与Rb基因表达率之间明显正相关 (L78:r =0 .743 ,P <0 .0 1;PG :r =0 .75 5 ,P <0 .0 1)。结论  9 cisRA可能通过Rb基因表达增加和CyclinD1基因表达减少途径使L78、PG细胞明显阻滞在G0 G1 期 ,并诱导肺癌细胞凋亡。

【Abstract】 Objective To investigate the relationship between 9-cis-retinoic acid-induced apoptosis of human lung carcinoma cell lines (L78 and PG) and expression of Cyclin D1,Rb gene proteins.Methods In vitro,L78 and PG cell lines were divided into two groups randomly:group A treated with 5 μmol/L 9-cis RA;group B treated with 0.1 % dimethylsulfoxid.After 48 h incubation,the apoptosis and Cyclin D1,Rb gene expression were detected by FACS techniques.Results In group A of the two cell lines,the apoptosis induced by 9-cis RA was increased significantly.There was a negative relationship between Cyclin D1 gene and the rate of cancer cell apotosis(L78:r=-0.707,P<0.01;PG:r =-0.785,P<0.05),and there was a positive relationship between Rb gene and the rate of cancer cell apotosis (L78:r=0.743,P<0.01;PG:r=0.755,P<0.01).Conclusion 9-cis RA can arrest cells in G 0/G 1 status by Rb up-regulation and Cyclin D1 down-regulation,then induce lung cancer cells apoptosis.
