雄激素对大鼠阴茎组织血管内皮生长因子及其受体表达的影响
Effect of androgen on VEGF and VEGFR-2(FLK-1/KDR) expression in rat corpus cavernous
【摘要】 目的 研究雄激素对大鼠阴茎组织中血管内皮生长因子 (VEGF)及其受体Flk 1 KDR表达的影响。方法 2个月龄SD雄性大鼠随机分成假手术组 (A组 ) ,去势组 (B组 ) ,去势 +睾酮替代组 (C组 ) ,以及 5α还原酶抑制剂保列治喂饲组 (D组 ) ,2周后取大鼠阴茎组织行VEGF及其受体Flk 1 KDR免疫组织化学染色和VEGFmRNA原位杂交 ,运用计算机图像分析系统 ,比较其表达强度。结果 A组和C组大鼠阴茎组织VEGF免疫组织化学染色均呈高表达灰度值( 99.80± 2 .88和 98.70± 3 .41) ,而B组与D组均呈低表达灰度值 ( 12 7.80± 3 .93和 12 5 .30±3 .76 ) ,(P <0 .0 1) ;VEGFmRNA原位杂交也显示B组 ( 15 9.30± 9.42 )与D组 ( 15 7.30± 9.71)阳性着色均明显少于A组 ( 12 4.6 0± 8.37)和C组 ( 12 5 .30± 7.80 ) ,(P <0 .0 1) ;而B组与D组之间 ,A组和C组之间表达灰度值差异均无显著性 ;VEGF受体Flk 1 KDR在各组表达差异均无显著性。结论 雄激素对大鼠阴茎组织中VEGF以及VEGFmRNA的表达均有上调作用 ,而对VEGF受体Flk 1 KDR的表达无影响 ;调节大鼠阴茎组织VEGF表达的雄激素活性成分可能主要为 5α 双氢睾酮。
【Abstract】 Objective To investigate the regulation of androgen on vascular endothelial growth factor(VEGF)and VEGFR-2(Flk-1/KDR) in rat corpus cavernous.Methods 40 two month old Male Sprague-Dawley rats were randomized into four groups:sham operated control(A),simple castration(B),castration in combination with testosterone supplementation (C) and sham operation plus 5alpha-reductase inhibitor finasteride(D).Two weeks later,all animals were sacrificed and infused with 4 % paraformaldehyde.Corpus cavernous were paraffin-embedded for sections.The expression of VEGF and Flk-1/KDR in corpus cavernous was detected by immunohistochemistry and VEGFmRNA by in situ hybridization techniques.The intensity of immunostaining was measured using image analysis system.Results Both Castration and 5alpha-reductase inhibitor finasteride administration caused a marked reduction in the expression of VEGF and VEGFmRNA in corpus cavernous.The expression of VEGF in groups B and D was 127.80±3.93 and 125.30±3.76,significantly lower than in group A (99.80± 2.88) and group C (98.70±3.41) (P<0.01).The expression of VEGF mRNA was also significantly lower in group B (159.30±9.42) and group D (157.30±9.71) than in group A (124.60±8.37) and group C (125.30±7.80) (P<0.01).There was no significant difference in both VEGF and VEGFmRNA expression between group B and group D or group A and group C.Different andrgen status had no effect on Flk-1/KDR expression.Conclusion Androgen can up-regulate the expression of VEGF but not VEGFR-2(Flk-1/KDR) in rat corpus cavernous;DHT seems to be the major androgen responsible for the expression of VEGF and VEGFmRNA.
