颈动脉粥样硬化性狭窄动物模型的建立
Establishment of an animal model of carotid atherosclerotic stenosis
【摘要】 目的探讨建立经济实用、稳定可靠的适于颈动脉粥样硬化性狭窄(CASS)外科治疗研究的CASS动物模型的条件。方法新西兰白兔25只,采用血管内膜气体干燥损伤法在动物的颈总动脉上造成特定条件的损伤,然后以特定高脂饲料喂养动物不同时间。评价各组动物血管狭窄程度和病理改变特点。结果1月组、2月组和3月组各组中重度狭窄均达到80%。病理检查证实高脂喂养2个月时,动脉的粥样硬化病理改变已属于较成熟的纤维斑块期。结论按本实验方法,损伤后喂养2个月的动物颈动脉狭窄程度和病理改变程度皆符合CASS外科治疗之实验研究需要。
【Abstract】 Objecti ve To establish the methods and factors affecting the development of an economica l,practical,stable and reliable animal model suitable for experimental studies of surgical treatments of carotid atherosclerotic stenosis(CASS).Methods Us ing25New Zealand rabbits,lesions were created by air-drying isolated segment s of carotid arteries(120ml ?min×15min)and feeding the rabbits a diet o f2%cholesterol,6%peanut oil for15days,1month,2months and3months sep arately.Digital subtraction angiography was used to evaluate the stenosis of ca rotid arteries.Many histo logical methods,such as hematoxylin staining瑂pecia l staining and several kinds of immunohistochemical analyses,were used to illus trate the characteristics of the atherosclerotic lesions,and then to screen out the best method to develop the CASS animal model.Results The average rates of stenosis were calcu lated and described as following,half-month group:46. 32%±19.22%;1-month group:72.80%±19.28%;2-month gro up:73.47%±22.26%;and3-month group:59.19%±25.22%. The percentages of moderate to severe stenosis of the4groups were all more than 80%,and the percentage of severe stenosis of the last3groups were closed to or more than50%.The pathological studies suggested that the atheroscle roti c lesions getting more and more severe as the feeding time went on.Fed the at herogenic diet for15days,the lesions fell into the fatty streak period of ath erosclerosis;fed for1month,the lesions belong to the early fibrous plaque per iod;fed for2month,the lesions fell into the mature fibrous plaque period;and fed for3month,the lesions were belong to the early atheromatous plaque period .Conclusion Injured and fed as described by the authors,the animal model of2-month group is stable and reliable,and it meets the demand of experimental st udies of surgical treatments of CASS.
