脑源性神经营养因子在激活态雪旺细胞中表达的初步实验研究
Primary experimental study of brain-derived neurotrophic factor(BDNF)expression in activated Schwann cell
【摘要】 目的 分析激活态雪旺细胞的脑源性神经营养因子 (brain -derivedneurotrophicfactor ,BDNF)表达及其对神经再生的作用。方法 选用 2 0只SD大鼠 ,将大鼠右侧正中神经及尺神经在腋部切断进行激活 (实验组 ) ,左侧的正常神经为 (对照组 )。采用双酶消化法获取雪旺细胞并进行激活后提取mRNA ,应用RT PCR的方法比较BDNFmRNA的变化。结果 激活态雪旺细胞BDNFmRNA表达比对照组明显增多。结论 激活态雪旺细胞分泌神经营养因子BDNF增多 ,具有促进神经再生的作用。
【Abstract】 Objective To ananlyse BDNF expression in activated Schwann cell and its effect on nerve regeneration. Methods The right median and ulnar nerves were transected at the axillary level for activation of Schwann cells (experimental group) in 20 SD rats, with their left median and ulnar nerves as control group. Schwann cells were obtained by double kinases digestion and mRNA was distilled from Schwann cells.RT PCR was used to compare the changes of BDNFmRNA. Results The expression of BDNFmRNA in activated Schwann cells is much more than that in the normal Schwann cells. Conclusion Activated Schwann cell improves the serection of BDNF which can promote nerve regeneration.
