汉坦病毒山东分离株的分子生物学研究
Molecular analysis of hantavirus isolated from Shandong province
【摘要】 目的 应用聚合酶链反应 (PCR)、核苷酸序列分析等方法 ,比较山东省家鼠型汉坦病毒的生物学特性。方法 提取汉坦病毒感染细胞的全细胞RNA ,逆转录后用聚合酶链反应扩增病毒cDNA。对PCR产物进行核苷酸序列测定。结果 汉坦病毒山东分离株SD2 5、SD70、SD2 7和SD10均为家鼠型汉坦病毒 ,核苷酸及氨基酸序列与已知SEO型病毒具有较高的同源性 ,这 4株病毒间的同源性高达 98%以上。结论 4株病毒分离时间跨度虽有十几年 ,但相互间变异很小 ,证实了家鼠型汉坦病毒的稳定性。
【Abstract】 Objective To compare the molecular characteristics of hantanviruses isolated from Shandong province by using PCR typing and nucleotide sequencing Methods Total cellular RNA was extracted from hantanviruses infected Vero E6 cells, viral cDNA was amplified by polymerase chain reaction PCR products of partial M segments of 4 strains of hantaviruses were sequenced Cross neutralization tests were performed Results Four strains of hantavirus isolated from Rottus in Shandong province were SEO like viruses The homology between Shandong isolates and other SEO like viruses was high at both amino acid and nucleotide levels The homology among those 4 strains of hantanviruses was 98% Conclusion The SEO like hantaviruses were more conserved than other hantaviruses The homology of SEO like hantaviruses isolated from Shandong province was as high as 98% at nucleotides level, though they were isolated at more than 10 year intervals
