半边旗5F对系统性硬皮病患者成纤维细胞增殖和胶原合成的影响

【摘要】 目的研究中草药半边旗5F对系统性硬皮病患者皮肤成纤维细胞的作用,以期找到治疗该病的新的有效药物。方法用四甲基偶氮唑盐(MTT)方法和光度检测法及逆转录-聚合酶链反应(RT-PCR)法测定半边旗5F对成纤维细胞增殖和培养基中的胶原含量以及Ⅰ、Ⅲ型前胶原mRNA表达的影响。结果半边旗5F对患者皮肤成纤维细胞的增殖具有显著的抑制作用,还能显著降低该细胞培养基中可溶性胶原的含量(P<0.01)。上述抑制作用随着药物浓度的增高和作用时间的延长而增强。采用RT-PCR法对Ⅰ、Ⅲ型前胶原的mRNA检测显示随着药物浓度的增高二者的表达量降低。结论半边旗5F对系统性硬皮病患者皮肤成纤维细胞的增殖和胶原合成具有显著的抑制作用,为寻找有效治疗该病的药物提供了新的启示。

【Abstract】 Objective To study the effect of herbal medicine Pteris semipinnata5F on the fibroblasts from systemic scleroderma(SSc)skin and explore new drug for SSc treatment.Methods The SSc skin fibroblasts were cultured as an experimental model and the effects of Pteris semipinnata5F were detected.The cell proliferation was determined by MTT assay,and the collagen production by photometric determination.The mRNA expression for typeⅠandⅢprocollagen was detected by RT-PCR.Results It was shown that Pteris semipinnata5F could not only significantly inhibit the proliferation of the cultured SSc skin fibroblasts,but also significantly reduce the soluble collagen content in the culture medium(P<0.01).The inhibitory effect was increased along with the increasing the concentration of the drug and prolonging the time of action.The results of RT-PCR showed the expression of typeⅠandⅢprocollagen was decreased along with the increasing of the concentration of the drug.Conclusions The above results demonstrate that Pteris semipinnata5F could significantly inhibit the proliferation and the collagen synthesis in cultured SSc skin fibroblasts.It may be developed as a new drug for the treatment of SSc.
