人血浆中右美沙芬及去甲右美沙芬的HPLC-荧光法测定

【摘要】 建立了简便、准确、灵敏的反相高效液相色谱 -荧光检测法测定人血浆中右美沙芬和活性代谢物去甲右美沙芬的血药浓度。线性范围分别为 0 .5~ 30 ng/ m l和 2 0~ 12 0 0 ng/ ml,定量限分别为 1.0和 2 0 .0 ng/ ml,回收率在97%以上 ,日间、日内 RSD小于 8.5 %。对 18名受试者单剂量口服 6 0 mg右美沙芬缓释混悬液后至 2 4 h血浆样品进行了测定 ,血浆中去甲右美沙芬浓度比右美沙芬高 10 0倍左右。

【Abstract】 A HPLC method with fluorescence detection for determination of dextromethorphan (1) and its active metabolite, dextrorphan (2) in human plasma was presented. The linear ranges of 1 and 2 were 0.5~30 ng/ml( r =0.9996) and 20~1200 ng/ml( r=0.9994 ), respectively. The limits of quantitation were 1.0 and 20.0 ng/ml, respectively. The average recovery was over 97%, and RSD s of intra- and inter-day were below 8.5%. In addition, the plasma concentrations over 24 h following single dose of 60 mg dextromethorphan hydrobromide were determined. The concentration of 2 was 100 times higher than that of 1.
