脂质体介导的聚集素反义寡核苷酸对膀胱癌EJ细胞的作用
Study of the effect of lipofectine-induced antisense oligonucleotide of clusterin on bladder cancer EJ cells
【摘要】 目的 探讨clusterin对膀胱癌EJ细胞生物学特征的影响。 方法 应用脂质体包裹的clusterin反义寡核苷酸转染EJ细胞 ,阻断其表达 ,通过 3′端末端标记、MTT试验、伊红排斥实验等方法观察clusterin反义基因对EJ细胞增殖、凋亡、坏死等生物学行为的影响。 结果 转染clusterin反义寡核苷酸组 (反义组 )EJ细胞凋亡增加 ,第 5天达峰 ,凋亡率 80 % ,与转染clusterin正义寡核苷酸组 (正义组 )比较 ,差异有显著性意义 (P <0 .0 5 )。反义组细胞死亡率增加 ,以转染后 1~ 4d明显 (13.8%~ 33.3% ) ,显著高于相应正义组 (P <0 .0 5 )。反义组细胞增殖活性降低 ,以转染后第 6、7天明显 ,显著高于相应正义组 (P <0 .0 5 )。 结论 clusterin是重要抗凋亡因子 ,其表达与膀胱癌细胞的生存、增殖、抗凋亡发展密切相关。
【Abstract】 Objective To investigate the effect of clusterin on the cytobiological features of bladder cancer EJ cells. Methods EJ cells were transfected using lipofectine-induced antisense oligonucleotide of clusterin,and their expression was blocked.The biological behaviors of EJ cells,such as proliferation,apoptosis and necrosis,were observed. Results Compared with the sense group which were transfected by sense oligonucleotide of clusterin,the antisense group which were transfected by antisense oligonucleotide of clusterin exprerienced increase in apoptosis of EJ cells,reaching the peak on day 5 with apoptosis rate of 80%.The difference between the 2 groups was statistically significant ( P <0.05).The cell death rate of the antisense group in increased,markedly on Days 1 to 4 after transfection(13.8%~33.3%),substantially higher than the sense group ( P <0.05).The cell proliferative activity of the antisense group decreased,markedly on Days 6,7,substantially loweer than the sense group( P <0.05)? Conclusions Clusterin is an important anti-apoptosis factor. The expression of clusterin is closely associated with the survival,proliferation and anti-apoptosis of the bladder cancer cells.
