【摘要】 目的　检测环氧化酶 2 (COX 2 )在大鼠睾丸组织中的表达情况 ,探讨COX 2在机体正常生殖机能中的作用机理。　方法　采用免疫组织化学、RT PCR和Western blot方法检测 5只正常成年大鼠睾丸组织中COX 2的表达。　结果　RT PCR和Western blot方法证实COX 2在基因和蛋白质水平均有表达 ;免疫组织化学结果显示 5只大鼠睾丸组织生精细胞中均有COX 2表达 ,其中初级精母细胞为强阳性 ,COX 2定位于曲细精管内的生精细胞。　结论　正常成年大鼠睾丸的生精细胞中存在COX 2表达 ,COX 2可能与睾丸的精子生成过程有关。更多还原

【Abstract】 Objective To confirm the expression and localization of COX-2 gene in mature rat testis,as COX-2 not only plays a central role in inflammation by up-regulating the synthesis of prostaglandins,but also mediates critical reproductive functions by other mechanisms. Methods In the mature Wistar rats,COX-2 expression and its localization were examined by using immunohistochemistry,RT-PCR and Western-blot. Results The results of RT-PCR and Western-blot showed that COX-2 mRNA and its protein exist in the testis. Expression of COX-2 was localized in spermatogenic cells on immunohistochemical study.No COX-2 expression in sertoli cells and leydig cells was found. Conclusions The present study confirmed the expression of COX-2 in rat testis and its localization in spermatogenic cells.These suggest that COX-2 may play a role in spermatogenesis.更多还原
