磷灰石纳米粒子体外致肝癌细胞的胀亡

【摘要】 [目的]研究磷灰石纳米粒子体外抗肝癌作用的机理。[方法]采用形态学观察和MTT法检测磷灰石纳米粒子作用于人肝癌细胞系Bel鄄7402的量效和时效关系。流式细胞仪分析细胞周期的时相变化。透射电镜下观察细胞超微结构的改变。[结果]磷灰石纳米粒子在体外对人肝癌细胞系Bel鄄7402具有明显的抑制作用,并呈现良好的量效和时效关系。肝癌细胞的生长阻滞于G1期。细胞体积增大,胞质空泡化,染色质凝集,胞浆溶解。[结论]磷灰石纳米粒子具有体外抗肝癌作用,细胞增殖阻滞于G1期,阻断细胞周期的进展,导致癌细胞胀亡。

【Abstract】 To explore the anti-cancer mechanism with apatite nanoparticles for hepatocellular carcinoma in vitro.The time and dose effect-dependent manners were investigated by the MTT method and morphology.The changes of cycle phase were analyzed by FCM and the cell ultrastructure was observed with TEM.Good time dependent efficacy and good dose dependent efficacy were revealed.Apatite nanoparticles revealed markedly in-hibitive effect to Bel-7402hepatocellular carcinoma cells.The proliferation of the human hepatocellular carcinoma cells was arrested at G 1 phase in cell cycle.Many changes were found such as cell enlarged,cytoplasm vacuoles,chro-matin agglutinated and cytoplasm dissolved.[Conclusions]Apatite nanoparticles have the efficacy of anti-hepatocellular carcinoma in vitro.The cell proliferation was arrested at G 1 phase and progress of the cell cycle was blocked.Those re-sult in the tumor cells oncosis.
