丙烯腈作业工人外周血有核细胞线粒体DNA损伤的研究

【摘要】 目的 探讨职业接触丙烯腈 (ACN)对工人潜在的衰老作用。方法 以多聚酶链反应(PCR)法测定接触ACN工人和对照组各 47人外周血有核细胞线粒体DNA(mtDNA)缺失率 ,同时 ,以老年人和青少年各 1 2名作对照研究。结果 mtDNA缺失片段扩增的阳性率 ,在职业接触丙烯腈的工人和老年人组中分别为 1 7.0 2 %和 2 5 .0 0 % ,对照组和青少年组未扩增出mtDNA缺失片段。结论 ACN可引起职业接触人群外周血有核细胞mtDNA的缺失 ;ACN对职业接触人群的衰老在分子水平上有相关的迹象

【Abstract】 Objective To study the potential aging effect on workers exposed to acrylonitrile(ACN). Methods The deletion rates of mitochondrial DNA(mtDNA) in peripheral blood nucleate cells of 47 exposed workers and 47 non exposed workers(as control),as well as 12 old people and 12 young people were measured with polymerase chain reaction(PCR). Results The positive rates of mtDNA deletion in peripheral blood nucleate cells were 17.02% in the workers exposed to ACN and 25.00% in group of old people.However,the mtDNA deletion was not detected in the control group and young people. Conclusions ACN could induce mtDNA deletion in peripheral blood nucleate cells of the exposed workers.There may be a potential molecular effect of occupational ACN exposure on workers’ aging.
