E-钙粘附素和DNA含量与胃癌侵袭、转移的相关性研究
Correlation of E-cadherin expression and DNA level with invasion and metastasis of gastric carcinoma
【摘要】 目的探讨E 钙粘附素 (E cadherin ,)在胃癌组织中的表达和癌细胞核DNA含量与胃癌侵袭、转移的相关性。方法应用免疫组织化学S P法对 83例胃癌组织、2 5例正常胃组织进行E cadherin表达的测定 ,同时以经典的Feulgen染色法及图象分析技术对癌细胞DNA含量进行测定 ,分析与胃癌侵袭转移之间的关系。结果正常胃粘膜E cadherin呈强阳性表达 ,胃癌组织中E cadherin表达率明显减低为 16 .9% (14 / 83)。E cadherin表达减低与胃癌细胞分化差、浸润深度、生长方式、淋巴结转移密切相关 (P <0 .0 5 ) ;而与TNM分期及肿瘤大小未见明显相关性。DNA指数在胃癌组织和正常胃组织中分别为 1.4 7± 0 .2 3和 0 .97± 0 .15 ,胃癌组织的DNA指数明显高于正常胃组织 (P <0 .0 1) ,DNA指数与胃癌的分化程度、浸润深度、生长方式、淋巴结转移相关 (P <0 .0 1) ,而与TNM分期和肿瘤大小无明显相关性。胃癌中E cadherin表达与DNA含量呈负相关。结论E cadherin表达和DNA含量与胃癌侵袭转移密切相关 ,二者在癌组织中水平的变化是胃癌细胞重要的恶性生物学特征 ,对其检测有助于对胃癌患者的转移及预后作出判断。
【Abstract】 Objective To evaluate the correlation between the expression of E-cadherin in gastric carcinoma, DNA level of cancer cells with invasion and metastasis of gastric carcinoma. Methods The expression of E-cadherin was observed in 83 cases with gastric cancer, 25 samples of normal stomach tissue by using streptavidin-peroxidase immunohistochemistry technique, and DNA level was quantitatively determined which was calculated by computer automatically with Feulgen staining in order to analyze its relationship with invasion and metastasis of gsatric carcinoma. Results E-cadherin in normal stomach tissues was strongly postively expressed , the expressing rate of which reduced to 16.9%(14/83) in gastric carcinoma tissue. The reduced expression of E-cadherin was closely related with lower differentiation, invasion depth, growth way and lymph node metastasis of gastric cancer cell(P<0.05); while there was no obvious correlation between TNM staging and tumor size. DNA indexes in gastric cancer tissue and normal gastric tissue were 1.47±0.23 and 0.97±0.15, respectively, suggesting that DNA index in gastric cancer tissue was obviously higher than that in normal gastric tissue(P<0.01). However, DNA index was correlated with differentiation degree, invasion degree, growth way and lymphnode metastasis of gastric cancer, which was not correlated with TNM staging and tumor size.E-cadherin expression in gastric cancer was negatively correlated with DNA level. Conclusion E-cadherin expression and DNA level are closely related with gastric cancer metastasis. The changes of E-cadherin and DNA levels are the important biologic character of gastric cancer cell, which help to judge the gastric cancer metastasis and the patients′ prognosis.
