多巴胺受体亚型D4和D5在人胃、十二指肠的定位及表达
Localization and mRNA expression of dopamine receptor subtype D4 and D5 in human stomach and duodenum
【摘要】 目的 探讨人体胃、十二指肠是否有多巴胺受体亚型D4和D5的表达及表达的部位。方法 人体胃、十二指肠标本液氮保存 ,低温固定 ,石蜡包埋 ,组织块连续切片 ;地高辛标记的寡核苷酸探针行原位杂交 ;病理图像分析系统进行分析并积分。结果 D4和D5在人体胃、十二指肠均有表达。在胃相对集中于腺体间质细胞及近黏膜肌层的黏膜固有层间质细胞上 ;而在十二指肠则较为弥散 ,除表达于十二指肠腺体间质细胞外 ,在黏膜肌层及布氏腺内亦有表达。D4和D5在胃和十二指肠的分布形态相似。D4在胃内的表达水平高于D5,阳性目标个数为 4 1.2 9± 5 .0 6比 2 6 .2 5± 5 .82 ;数密度为 4 .6 8± 0 .5 9比 2 .97± 0 .6 5 ,差异均有显著性 (P值均 <0 .0 1) ;在十二指肠黏膜内阳性目标个数为 30 .71± 10 .0 6比 2 2 .18± 4 .96 ,数密度为 3.4 8± 1.13比 2 .5 2± 0 .5 7,差异均有显著性 (P值均 <0 .0 1)。D4在胃内的表达高于十二指肠 ,D5表达无差异。结论 D4和D5在人体胃、十二指肠均有分布 ,主要位于腺体间质细胞上 ,十二指肠黏膜肌层及布氏腺亦有少量分布 ;D4的含量高于D5,且以胃内表达最高。
【Abstract】 Objective To investigate the localization and mRNA expression of dopamine receptor subtype D4 and D5 mRNA in human stomach and duodenum by in situ hybridization. Methods The oligonucleotide probe was synthesized and labeled with digoxin, and then hybridized with human gastric and duodenal samples, which were stored in liquid nitrogen, fixed in hypothermia, and embedded in paraffin. The results were recorded and quantitatively analyzed using the pathological image computer analysis system. Results Dopamine receptor subtype D4 and D5 mRNA both expressed in human stomach as well as duodenum. They distributed mainly in the interstitial of mucosal glands and the lamina propria near lamina muscularis mucosa. The mRNA expression value of D4 was higher than that of D5 in both stomach and duodenum. In stomach, the number of positive target cells was 41.29±5.06 vs. 26.25±5.82 ( P < 0.01 ) and the density was 4.68± 0.59 vs. 2.97±0.65 ( P <0.01); similarly, in duodenum, the number of positive target cells was 30.71±10.06 vs. 22.18±4.96 ( P <0.01) and density was 3.48± 1.13 vs. 2.52±0.57 ( P <0.01). D4 mRNA expression in stomach was higher than that in duodenum( P <0.01). Conclusions Dopamine receptor subtype D4 and D5 can both express in human stomach as well as duodenum. The value of D4 mRNA expression may be higher than that of D5 in both stomach and duodenum. The values of D4 and D5 mRNA expression may be higher in stomach than in duodenum respectively.
