树突状细胞和巨噬细胞对肝癌细胞抗原的提呈作用

【摘要】 目的 比较同一个体来源的树突状细胞(DC)和巨噬细胞(MΦ)对肝癌细胞抗原的提呈作用。 方法 从同一健康外周血中分离出单核细胞,在粒细胞-巨噬细胞集落刺激因子(GM-CSF)联合白细胞介素-4(IL-4)或单独在GM-CSF的作用下,分别诱导出DC和MΦ;DC或MΦ与自体淋巴细胞和肝癌细胞抗原共培养5d,结束培养前12 h加入37 kBq/孔(1 μci=37 kBq)3H-胸腺嘧啶,收获细胞,用液体闪烁计数仪测定增殖效应(cmp值)。 结果 接触肝癌细胞抗原的DC和MΦ都具有显著地刺激自体淋巴细胞增殖的能力,而以DC的刺激能力更为明显。加20μl/ml抗原时刺激效应最明显,DC组、MΦ组和淋巴细胞组的cpm值分别是11950.3±1621.8、8708.5±176.1和402.5±43.1。结论 同一个体来源的DC的抗原提呈能力明显强于MΦ。

【Abstract】 Objective To study the role of dendritic cells (DCs) and macrophages, differentiated from the same individual peripheral blood monocytes, in tumor antigen- presenting. Methods DCs and macrophages were differentiated from human peripheral blood monocytes by adding both Granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-4 (IL-4) or GM-CSF only. Then they were loaded with tumor antigen at different concentrations and cocultured with autologous T cells in 96-well flat-bottomed microtiter plates for five days at 37℃, 5% CO2. 3H-thymine was added before the culture terminated, and twelve hours later, the cells were gathered to test the cpm value. Result Both DCs and macrophages chased with tumor antigen could strongly stimulate the proliferation of autologous T cells, especially DCs. The stimulation effect with 20 μl/ml antigen was the most remarkable and the cmp values were 11 950.3±1 621.8, 8 708.5±176.1, 402.5±43.1 in DCs group, Macrophages group, and lymphocytes group, respectively. Conclusion The antigen presenting role of DCs is stronger than that of macrophages from the same individual.
