HPLC法测定甘肃产小檗属植物不同部位的生物碱
Determination of Four Alkaloids in Berberis Plants By HPLC
【摘要】 目的 :建立同时测定三颗针植物不同部位小檗碱、药根碱、巴马汀、小檗胺含量的HPLC法 ,并进一步考察 11种甘肃产小檗属植物不同部位生物碱的分布状态。方法 :检测波长小檗碱、药根碱、巴马汀为 346nm ,小檗胺 2 12nm。流动相乙腈 0 .0 2mol·L-1磷酸二氢钾水溶液 ,梯度条件 0~ 8min ,乙腈 32 %~ 36 % ;8~ 2 0min ,36 %~ 5 1%。流速1.0mL·min-1。结果 :小檗碱、药根碱、巴马汀、小檗胺的线性方程、相关系数、线性范围分别为 :Y =4 .0 74× 10 5X+80 80 ,r=0 .9998,0 .0 2 8~ 4 .74 μg ;Y =4 .5 88× 10 6X +16 780 ,r =0 .9996 ,0 .0 12~ 2 .0 μg ;Y =3.74 3× 10 6X +2 12 4 7,r =0 .9999,0 .0 2 6~ 0 .5 2 μg ;Y =4 .4 73× 10 6X +36 342 0 ,r =0 .9998,0 .0 15~ 2 .5 6 μg。 结论 :不同产地、不同品种、不同部位三颗针植物中生物碱的含量分布有明显的差异。
【Abstract】 Objective: In order to assess Berberis , a simple and effective high-performance liquid chromatographic (HPLC) method was established. Method: By applying HPLC with gradiation elution, this method was developed for determination of four kinds of alkaloids including berbamine, Jatrorrhizine, berberine and palmatine in methanolic extracts from root bark, root,stem bark and stem of Berberis . Result: The alkaloids berbamine, jatrorrhizine, berberine and palmatine shown good linear correlations in the range of 0.028~4.74 μg ( r =0.999 8);0.012~2.0 μg( r =0.999 6);0.026~0.52 μg( r =0.999 9);0.015~2.56 μg( r =0.999 8), respectively. The correlation coefficients of the calibration curve for the analytes exceeded 0.999 8. Conlusion:The optimized HPLC method was applied to analyze various samples, and the results shown that the content of alkaloids were obvious difference in Berberis from different area, different species and different parts.
