猪苓多糖对膀胱癌细胞癌基因蛋白表达的影响
The Expression of P53 and H-ras in Bladder Cancer Cells Induced by Polyporus Polysaccharide
【摘要】 目的:分析猪苓多糖对体外培养T24膀胱癌细胞癌基因蛋白表达的影响。方法:应用免疫荧光法、激光共聚焦显微镜对p53、H-ras基因蛋白表达进行定位、定量观察。结果:猪苓多糖显著增加p53蛋白表达,24小时表达最高,呈弥漫性分布,随后逐渐下降;对照组在24小时表达很弱,48小时表达最强。猪苓多糖对H-ras基因蛋白表达无明显影响。结论:猪苓多糖对T24细胞p53基因蛋白表达有一定的调节作用,对H-ras基因蛋白表达无明显影响。
【Abstract】 Objective:To elucidate the specific mechanisms of the expression of P53and H-ras in bladder cancer cells induced by polyporus polysaccharide(PPS).Methods:Laser scan-ning confocal microscope was used to investigate the distribution of the p53?H-ras protein in t24bladder cancer cells.Results:p53labeled with fluorescence spread all over the cytosol and nuclei in the T24cells after24h treatment with PPS,while the fluorescent intensity decreased after48h stimulation;No correlation with expression of H-ras in T24cells was found.Conclu-sion:These results suggested enhanced expression of P53of bladder cancer cells induced by PPS.
