穴位埋线加灸对衰老模型鼠抗自由基氧化损伤DNA作用的实验研究
Effect of point thread embedding plus moxibustion on DNA oxidative damage induced by free radicals in aging model mice
【摘要】 目的:探讨穴位埋线加灸延缓衰老的疗效及其可能机理。方法:利用小鼠吸入臭氧(O3)制造衰老模型。在其造模过程中,实施穴位埋线加灸治疗,9周后检测小鼠全血超氧化物歧化酶(SOD)的活性,肝组织丙二醛(MDA)的含量,并用彗星实验(comet assay)检测肝脾组织细胞DNA氧化损伤的程度。结果:经穴位埋线加灸治疗后,与模型组相比,小鼠全血SOD活性显著提高(P<0.05),肝组织MDA含量显著降低(P<0.01);脾细胞DNA拖尾率显著下降(P<0.05),拖尾尾长显著减短(P<0.01);但肝细胞DNA损伤的拖尾率和拖尾尾长无显著变化(P>0.05,P>0.05)。结论:穴位埋线加灸能提高机体抗自由基氧化损伤的能力,减轻脾细胞DNA损伤程度,达到延缓衰老的目的,但其作用的组织器官存在一定的特异性。
【Abstract】 Objective To observe effects of point thread embedding plus moxibustion on dekying aging process and explore the mechanism. Methods Aging model mice were established by inspiration of ozone (Qs). During the course of establishment of the model mouse, point thread embedding plus moxibustion was applied for 9 weeks and then the activity of blood superoxide dismutase (SOD) and malondiadehyde (MDA) content in liver were determined, and the oxidative damage degrees of DNA in the liver and spleen were detected by comet assay. Results In the point thread embedding plus moxibustion group the blood SOD activity increased significantly (P<0.05), MDA level in the liver and the comet rate of spleen DNA decreased significantly (P<0.01, P < 0.05), and the comet length of spleen DNA was shortened significantly (P<0.01) as compared with the aging model group. However, the comet rate and comet length of liver DNA had no significant changes (P >0.05). Conclusion The treatment of point thread embedding plus moxibustion can increase the ability of protecting the organism from oxidative damage of free radicals and reduce the damage degree of spleen DNA, so as to delay aging. And the action is of specificity for tissues and organs.
